The Franquero Dynamics Framework (FDF) v2.1: System and Methods for Quantitative Systems Pharmacology and Temporal Optimization of Multi Agent Immuno Oncology Therapies via Dual Axis Glycocalyx Modification
By Keny Wayne Franquero.
Abstract
This preprint outlines the mathematical and biophysical specification for the Franquero Dynamics Framework (FDF) v2.1. The architecture implements a 23-state ordinary differential equation (ODE) modeling system, termed BASET (Barrier, Access, Signal, Effector, Toxicity), to analyze quantitative systems pharmacology in immuno oncology. Key innovations include a dual axis glycocalyx modification model separating grafting density (G) scission from backbone contour length (Lc) truncation, a q72h dose fractionation protocol that doubles integrated access AUC, and rigorous off target selectivity bounds selₑₙ₂ ≤ 0.005 decoupling efficacy from vascular leak toxicity.
