Abstract
Genetic variations in folate metabolism-related genes have been proposed as potential risk factors for carcinogenesis. This investigation aimed to evaluate the association between the SLC19A1 (C2522T) polymorphism and the risk of colorectal cancer (CRC). This study was conducted on 100 CRC patients and 100 age- and gender-matched healthy individuals as a control group. Genotyping of SLC19A1 (C2522T) gene polymorphism was made by tetra-allele refractory mutation system (T-ARMS) PCR method. Patients and controls were comparable regarding age, gender, or smoking status. Regarding SLC19A1 (C2522T), the CT genotype was more frequent among CRC patients compared to controls, however the variation was not statistically significant. Similarly, the T allele frequency was slightly higher in CRC patients than controls, but without statistical significance. After adjustment for age, gender, and smoking status, none of the codominant, dominant, recessive, overdominant, or log-additive models showed a substantial association with CRC risk. In conclusion, the present study demonstrated that the SLC19A1 (C2522T) polymorphism was not significantly related to the risk of CRC in the studied population. Further large-scale studies involving different ethnic groups and additional folate pathway genes are warranted to clarify the genetic determinants of CRC susceptibility.
