Abstract
The aim of the research was to study the functional parameters of the cardiovascular system in rats with experimental type 2 diabetes mellitus and with the administration of L-arginine and N-acetyl-L-cysteine. Materials and methods. Rats with diabetes mellitus type 2 (DM2) were initially induced with insulin resistance (high-fat feed for 8 weeks), after which they were administered a single dose of 30 mg/kg of streptozotocin. After 6 weeks, the animals were divided into 3 subgroups: without the administration of amino acids, rats with L-arginine and with N-acetyl-L-cysteine. To achieve the aim, mean blood pressure was measured and echocardiography was performed at weeks 0, 6, and 8 of the experiment. Results. The research showed a significant increase in blood pressure and the development of left ventricular myocardial hypertrophy in rats with DM2. An enlargement of the left ventricle cavity and impaired systolic function were also found. These changes progressed throughout the experiment. L-arginine contributed to a statistically significant decrease in blood pressure and an improvement in certain echocardiographic parameters. N-acetyl-L-cysteine also had a positive effect on the mass of the left ventricle, but to a lesser extent. Neither substance was able to fully restore the parameters to control values. Conclusions. Against the development of experimental type 2 diabetes mellitus in rats at week 6 left ventricle hypertrophy with dilatation and systolic dysfunction develops, accompanied by an increase in blood pressure. At week 8 of the experiment, rats without amino acids showed a progression of hypertrophy, dilatation and systolic dysfunction compared to week 6, but the mean arterial pressure remained unchanged. The use of the amino acids L-arginine and N-acetyl-L-cysteine has a positive effect on cardiohemodynamic parameters and blood pressure in type 2 diabetes mellitus, however, complete restoration to the physiological norm was not achieved, and the effect was more pronounced with L-arginine.
