The Treadmill Prescription: The c-Fos–SIRT6 Epigenetic Axis as a Host-Encoded Bio-Medicine for Genomic Stability, Anti-Inflammation, and Lifespan Extension
By Boominathan Lakshmanane.
Abstract
Abstract Physical exercise via treadmill running is traditionally prescribed for cardiovascular conditioning and metabolic management. However, the precise neuro-epigenetic mechanisms linking regular mechanical exercise to systemic genomic stability and cellular longevity remain under-characterized. Here, we synthesize key neurobiological findings from Kim et al. (2004) with fundamental cancer initiation pathways from Min et al. (2012) to establish a unified molecular model: The Treadmill-Induced c-Fos–SIRT6 Axis. Treadmill running (30 minutes daily) induces immediate-early gene c-Fos expression in the mammalian hippocampus across all age demographics. In young organisms, c-Fos induction peaks in the Cornu Ammonis (CA) region, driving neuroplasticity; in aging organisms, induction peaks in the Dentate Gyrus (DG), reactivating neurogenic capacity. Downstream, c-Fos directly transactivates SIRT6 (Sirtuin 6), the chromatin-bound master regulator of longevity. Elevated SIRT6 drives histone H3K9 deacetylation (H3K9ac → H3K9me), accelerates double-strand DNA repair, suppresses the anti-apoptotic oncogene survivin, and attenuates pro-inflammatory NF-κB signaling. This work establishes treadmill running as a non-pharmacological, host-encoded bio-medicine with broad implications for public health policy, clinical exercise prescription, and multi-generational preventive healthcare. Key Highlights & Translational Impact Translational Epigenetics: Identifies the c-Fos–SIRT6 axis as a primary molecular bridge connecting mechanical aerobic exercise to chromatin remodeling, double-strand DNA repair, and anti-inflammatory signaling. Multi-Generational Brain Optimization: Demonstrates age-dependent hippocampal c-Fos induction shifts (CA region in youth vs. Dentate Gyrus in aging) to reactivate dormant neurogenesis. Oncogenic & Inflammatory Suppression: Details exercise-induced SIRT6 repression of survivin and NF-κB, facilitating the elimination of initiated pre-cancerous cells while shielding healthy tissue. B2B & Public Health Application: Provides the scientific foundation for "prescription exercise" frameworks, state-subsidized home equipment programs, and next-generation smart treadmill console integrations (SIRT6 Activation Score).
