Abstract / Summary
Abstract Plants activate defense mechanisms with reactive oxygen species (ROS) and antioxidant enzymes in response to pathogen infection. The objective was to adapt the infiltration protocol, to detect ROS, using the dye nitrobluetetrazolium (NBT) in root tissues of bell pepper plants. The susceptibility of this genotype was determined in an incubator and the in situ localization protocol of ROS adapted by decreasing the NBT infiltration period without vacuum and addition of incubation time without light. Gall indices, egg masses, reproduction factor and tests to locate ROS were obtained 60 days after nematode inoculation. Meloidogyne enterolobii reproduced in the susceptible genotype cv. ‘Beth-R’. The protocol adaptation allowed to efficiently determine the process and extension of the M. enterolobii infection based in the increase of the gall area and formation of free radicals.
