Transforming Outcomes in Metastatic Uveal Melanoma: First Real‑World Experience with Tebentafusp in Slovenia
By Reberšek M, Studen A and Hribernik N.
Abstract
Background: Uveal melanoma is a rare cancer with poor prognosis. Tebentafusp is the first systemic therapy shown to improve outcomes and prolong survival in patients with metastatic uveal melanoma. Here, we present the first Slovenian real‑world experience with tebentafusp in metastatic uveal melanoma. Patients and Methods In this retrospective, observational study, we included metastatic uveal melanoma patients with the human leukocyte antigen A*02:01 genotype who received at least one infusion of first‑line systemic treatment with tebentafusp between April 2023 and December 2025 at the Institute of Oncology Ljubljana, Slovenia, with a data cut-off date for follow-up on June 30, 2026. Data were collected retrospectively and comprised baseline patient and disease characteristics, tebentafusp administration, response rates, toxicity, and survival outcomes. Results Of 16 screened patients, 9 were HLA‑A*02:01 positive. Two patients had rapidly progressive disease and died before receiving systemic or other treatment. A total of 7 patients were included in the final analysis. The median follow‑up was 19.9 months. The best overall response was stable disease. Median progression‑free survival was 10.5 months (95% CI, 4.7–not reached). Median overall survival was not reached (95% CI, 9.0–not reached). All 7 patients experienced treatment‑related adverse events, including cytokine release syndrome, all of grade 1–2. No treatment‑related deaths occurred. Conclusion In this real‑world retrospective study of metastatic uveal melanoma, tebentafusp was associated with outcomes comparable to those reported in the registrational phase III trial and was safely administered in daily clinical practice at experienced centres.
