Résumé
This research concept proposes a safety-gated nanothrombolysis platform in which a biocompatible nanoparticle carries a fibrinolytic payload and targets thrombus-associated features such as fibrin or activated platelets. Optional trace radionuclide labeling supports quantitative PET/SPECT biodistribution, localization, clearance and treatment-response assessment. The primary lytic mechanism is biochemical fibrinolysis; direct radiation-mediated clot dissolution is not assumed. The framework defines preclinical efficacy endpoints, bleeding limits, organ dosimetry, hematologic and biochemical toxicity measurements, nanoparticle clearance and stopping rules. Therapeutic and imaging functions require independent evaluation. No established clinical therapy, fixed human dose or demonstrated clinical efficacy is claimed. Expanded illustrated research concept paper, October 2026.
